# Cells use a little-known molecule to protect themselves from iron overload

## Summary

Researchers at MIT and the Whitehead Institute discovered that polyamines, small molecules abundant inside cells, act as storage lockers that keep iron in a non-reactive, safe state, preventing toxic iron overload. Using a genome-wide screen and a newly developed fluorescent sensor for reactive iron, the team found that when polyamine levels drop, cells depend on the protein GPX4 to survive, and reactive iron levels rise. The findings suggest combining polyamine-lowering drugs with GPX4 inhibitors could improve cancer treatment, and may also explain the elevated brain iron levels observed in early-onset Parkinson's disease linked to polyamine transport mutations.
